چکیده
An assay system for the in vitro transcription of the lac operon is described. A protein factor (CAP) and cyclic AMP, which are essential for the lac expression in vivo, also stimulate lac transcription in vitro.

Role of cyclic adenosine 3',5'-monophosphate and the cyclic adenosine 3',5'-monophosphate receptor protein in the initiation of lac transcription.
Optimal transcription of the lac operon in a defined purified in vitro system is dependent upon the concentrations of cyclic adenosine 3’,5’-monophosphate (cyclic AMP), the cyclic AMP-binding protein (CRP) and RNA polymerase. The concentration of CRP required for maximal Zac transcription varies proportionately with the concentration of lac DNA used as template. Maximal Zac transcription is obt...
متن کاملDifferential binding of cyclic adenosine 3' ,5'-monophosphate to the cyclic adenosine 3' ,5'-monophosphate receptor protein in Escherichia coli.
Binding of cyclic adenosine 3' ,5'-monophosphate (cAMP) by the cAMP receptor protein in crude cell-free extracts of Escherichia coli was characterized. When cell were grown in glucose, binding was inhibited 50% relative to extracts from cells grown with succinate as carbon source . This inhibition could be relieved by dialysis.
متن کاملRegulation of c-jun gene expression by cAMP in HL-60 myeloid leukemia cells.
Previous studies have demonstrated that expression of the c-jun proto-oncogene is induced by phorbol esters and other agents that activate protein kinase C. The present work has examined the involvement of cAMP-dependent signaling mechanisms in the regulation of c-jun gene expression. Low levels of c-jun transcripts were detectable in untreated HL-60 myeloid leukemia cells. In contrast, treatme...
متن کاملMechanism of activation by adenosine 3':5'-cyclic monophosphate of a protein phosphokinase from rabbit reticulocytes.
Two protein phosphokinases (EC 2.7.1.37) were found to be present in rabbit reticulocytes. The two enzymes were separated by DEAE-cellulose chromatography and called kinases I and II. Adenosien 3':5'-cyclic monophosphate stimulated the activity of both enzymes. However, the degree of stimulation was different and depended on the protein acceptor used. In the presence of adenosine 3':5'-cyclic m...
متن کاملDiscovery of a cAMP deaminase that quenches cyclic AMP-dependent regulation.
An enzyme of unknown function within the amidohydrolase superfamily was discovered to catalyze the hydrolysis of the universal second messenger, cyclic-3',5'-adenosine monophosphate (cAMP). The enzyme, which we have named CadD, is encoded by the human pathogenic bacterium Leptospira interrogans. Although CadD is annotated as an adenosine deaminase, the protein specifically deaminates cAMP to cy...
